Bob Li, MD, PhD, MPH, Memorial Sloan Kettering Cancer luca d'angelo Center, New York, NY, discusses the results of the Phase II CodeBreak 100 trial (NCT03600883) travis kelce of sotorasib (AMG 510) in KRAS p.G12C mutated non-small cell lung cancer (NSCLC). Sotorasib is a first-in-class irreversible inhibitor of KRAS-G12C, which showed early signs of efficacy and tolerability in the Phase I CodeBreak 100 trial. In total, 126 patients with advanced or pliskova metastatic KRAS p.G12C-mutant NSCLC were enrolled and followed for a median of 9.3 months. The results showed that sotorasib provided deep and durable responses, with an overall response rate of over 37% The treatment was well tolerated and has the potential to be the first targeted treatment option for this patient population in which outcomes are very poor.
